My name is Nadia. I was watching your webinar, titled “How to improve your results with Masterplex QT” with Allen Liu. I have a question about weighting. If the weighting does not improve my data’s % recovery; can I choose not to use weighting and still have solid data?

Dear Nadia,
I would incorporate weighting regardless because it will still affect the calculation of the parameters. The 4PL and 5PL assumes that you have homoscedastic data which, of course, is untrue.

Dear Mr. Liu,
Another quick question. If for example we have duplicates on standard wells, one of the wells has %recovery of 142 (above 130; not quite acceptable from my understanding) and the other one has %recovery of 84. Is this particular standard is usable (in terms of their mean %recovery is 113)?

Hi Nadia,
I have seen many different protocols so I really cannot say that what is right or what is wrong. I commonly see protocols where only points that are within 70-130% recovery are accepted and any replicates outside of this range are marked as outliers.
